The story centers around a 17-year-old high school delinquent Seiji Sawamura, who fights so much his right hand is known as "Demon Right Hand". This causes many people to be scared of him, and eventually believes his right hand is his only companion. That is until he wakes up one day to find a girl- Midori Kasugano- someone who claims to have loved him from a far for a long time, in place of  his right hand.

Sawamura Seiji the male protagonist of the story is a 17 year old highschool delinquent who loves to fight. He beats up angsters with his trusty "Devil's Righthand" and formally was known as "Mad Dog Sawamura" (This is however ironic due to his fear of dogs, which mentioned in the manga, was caused by his sister). Though Misunderstood, Sawamura only fights for the weaks and in facts hates bullying them. He is feared by everyone (except for his junior Osamu Miyahara and later by Ayase Takako who falls in love with him after being saved) which is why he never had a girlfriend. And one fateful day, just as when he lost all his hope of ever finding a girlfriend, Midori appears and replaces his right hand.

Midori, the female protagonist of the story is a 16-year-old student in an elite school. She had a crush on Sawamura for 3 years and her only wish was that she wants to be with seiji. And one morning, for unexplainable reasons she woke up as sawamura's right hand Midori is a fun, carefree girl, but as time progresses it is shown that she is actually a little bit more shy than she lets on (she mostly doesn't show it because she has been spending so much time with Seiji). Some emotions she hides well are her jealousy towards the other girls Seiji comes in contact with and her determination to win his heart. According to the manga, she seems to possess Herculean strength when she is angry or determined. Also, she seems to be able to drag Seiji around with ease as long as he isn't resisting (i.e. in a sleeping or unconscious statue). This might be because she possessed Seiji's "Devil right-hand", or simply because usual traits of anime girls (getting very violent when emotionally disturbed). Despite being born and raised from a well off family, Midori with the help of Makie, their house maid have talents like Cooking and sewing. she also loves to clean and she is very tidy.

Takako Ayase is a major character in Midori Days, and the Deutagonist. She is a high school girl who attends classes with Seiji Sawamura, and becomes smitten with him after being saved. Out of all the other characters in both the anime and manga, Takako gets a large amount of stories dedicated to her, or garners lots of screen time and development.
